Kalvanin Kadhali narrates the tale of Muththaiyan and Kalyani, a dacoit and his beloved. Kalyani, residing in PoonkuLam village, falls in love with the kind-hearted Muththaiyan. Despite circumstances labeling Muththaiyan as a dacoit, Kalyani remains devoted to him. Fleeing from the authorities, Muththaiyan lives as a fugitive in the forest. Eventually, they plan to escape together and start a new life far away. Muththaiyan's loyal friend, Kamalapathi, aids their getaway by disguising himself as a woman to communicate the plan to Muththaiyan. However, when Kalyani arrives at the rendezvous point, she misunderstands Muththaiyan's gratitude towards Kamalapathi as infidelity and leaves in anger. Inspector Shastri, in disguise, approaches Kalyani for Muththaiyan's whereabouts, and in her fury, she inadvertently reveals his location. Realizing Shastri's true identity, Kalyani chases after him. The tragic conclusion sees the lovers reuniting in death.
